Group A will feature Harimau Muda, Woodlands Wellington, Home United and National Football League team Admiralty FC.
Group C will see Albirex Niigata, Hougang United and Geylang International slugging it out for two slots in the quarter-finals.
Defending League Cup champions Brunei DPMM will face Balestier Khalsa and Warriors FC in Group D.
The tournament will be played from June 5 to June 23 with the final scheduled for the Jalan Besar Stadium.
